添加含有高浓度乙醇的红曲酒至福建传统红曲醋醋母中富集产酸菌株,采用高浓度乙醇平板,依据溶解圈指标从福建传统红曲醋液体循环工艺样品中分离出7株产酸菌株。综合菌株形态、生理生化实验以及16S r DNA序列测定等信息,确定这7株菌株分类地位为变形菌门(Proteobacteria)α-变形菌纲(Alphaproteobacteria)红螺菌目(Rhodospirillales)醋酸菌科(Acetobacteraceae)葡糖酸醋杆菌属(Gluconacetobacter),其中菌株Y5052、Y5054、Y5072、Y5092鉴定为斯氏葡糖酸醋杆菌(Gluconacetobacter swingsii);菌株Y5032、Y5033、Y5071鉴定为欧洲葡糖酸醋杆菌(Gluconacetobacter europaeus)。测定这些菌株的产酸能力,菌株Y5052产酸量最低,7 d达15 g/L;菌株Y5054产酸能力最强,7 d产酸量达57.0 g/L。  相似文献

It was recently demonstrated that the pineal neurohormone melatonin is a hydroxyl radical scavenger and antioxidant, and that it plays an important role in the immune system. In studies reported herein, we have investigated the relationship of the melatonin level and the NF-κ B DNA binding activity in the spleen of Sprague—Dawley rats. These in vivo results indicate that NF- κB DNA binding activity in the spleen is lower at night, when endogenous melatonin levels are elevated, than during the day, when endogenous melatonin levels are lower. Furthermore, exogenously administered melatonin (10mg/kg) was shown to cause a significant decrease in NF-κB DNA binding activity in the spleen at 60min after intraperitoneal injection (as compared with vehicle-treated rats). These new findings suggest that the normal night time rise which can be expected for melatonin may be associated with increased NF-κB DNA binding activity in the spleen. The melatonin, therefore, could potentially act to modulate spleen function and/or the immune system by regulating the NF-κB DNA binding activity in the spleen.  相似文献

Full grown oocytes derived from Bufo Bufo gargarizans rearing at high temperature environment (24℃), never underwent GVBD after progesterone treatment.No p34^cdc2 Hl kinase activity was detected in the oocytes after progesterone stimulation or OA microinjection;Western blotting analysis showed that the level of p34^cdc2 and p33 in the oocytes are significantly lower than those in the oocytes derived from the hibernating toads (below 10℃).^35S-Met incorporation analysis showed that when the oocytes were incubated at 6℃,synthesis of about thirty defferent polypeptides was promoted or induced,including p34^cdc2 and some other p13^suc1-binding proteins.All these results indicated that a low temperature environment is essential for the oocytes of Bufo Bufo gargarizans to express and stord some cell cycle drivers and its regulators,and to gain the maturation competence.These results will also provide a nwe clue for explaining the molecular mechanisms why gametogenesis of some organisms depends on a relative low temperature and how to maintain the geographical distribution of some animals.  相似文献

Shell-less Discinisca larvae of 2–3 p.c. (pairs of cirri)and small shelled larvae of 4 p.c. stages, hitherto undescribed,form a growth series with those previously described. The shellfirst formed during early 3 p.c. or early 4 p.c. stage. In swimmingthese young larvae did not rotate about their longitudinal bodyaxis, unlike larger larvae. In some larvae pigment granulesaggregated in the anterolateral stomach wall, forming "eye spots,"which are not comparable to the sensory eye spots of articulatelarvae. The order of appearance of embryonic setae and larvalsetae was described. The role of the former in floatation andin protective response was suggested. In recent brachiopod ontogeny there is an evolutionary simplificationfrom the presumably primitive condition in lingulids with shelledembryo, shelled larva with statocysts, long planktotrophic existenceand well developed trocholophe with continuous budding of cirrito 8-20 pairs; to the discinids with setiferous, shell-lessembryo, shelled larva with statocysts, shorter planktotrophicexistence and larval trocholophe with a maximum of 4 cirruspairs; and finally to the articulates with setiferous, shell–lessembryo and larva with no statocysts, no differentiated cirriand short free-swimming existence.  相似文献

白桦雌花从开花到雌性器官的成熟需经历1个月左右的时间,解剖学观察表明:四月下旬越冬的雌蕊原基开始了活跃的分裂和分化。子房和柱头开始生长。四月末开花,五月初授粉。此后胚珠开始长大。五月中旬即分化形成珠被,珠心,珠被为单层珠被,胚珠为厚珠心胚珠,胚珠倒生,五月中下旬,珠心内产生大孢子母细胞,一周左右发育为成熟胚囊-七细胞八核胚囊,五月末完成双受精,白桦胚胎发育经过合子,原胚,球形胚,心形胚和鱼雷形胚等时期最后发育成熟,胚乳发育与胚胎同步,即受精的极核进行几次分裂后形成核型胚乳,胚乳核不断增多,在形成心形胚后胚乳细胞形成细胞壁。  相似文献

为探讨水牛胚胎移植的效果 ,于 2 0 0 2年对云南水牛进行了胚胎移植试验 :①用国产氯前列烯醇(PG) 0 6mg/头·次处理供、受体水牛的同期发情率和可用率分别为 4 3 33% (13/ 30 )和 16 6 7% (5 / 30 ) ;同期发情率经产水牛高于青年水牛 (P =0 0 86 ) ,杂交水牛高于德宏水牛 (P =0 15 3) ,体重 4 0 1~ 5 30kg水牛显著高于体重 30 0~ 4 0 0kg水牛 (P <0 0 5 ) ;发情明显水牛的可用率极显著高于发情不明显的水牛 (P <0 0 1)。②选用河流型摩拉水牛与沼泽型德宏水牛的杂交一代 5头为供体 ,分进口激素组 (n =2 )和国产激素组 (n =3)进行超数排卵 ,共有 2头获 9枚胚胎 ;进口激素组供体的平均获胚数和可移植胚数分别为 2 0枚和 1 5枚 ,比国产激素组分别多 0 33枚 (P =0 4 5 4 )和 1 17枚 (P =0 2 88)。③所获 4枚可用鲜胚分别移植 3头受体 ,结果 90d的妊娠率为 33 33% ,但最终无一头产犊。试验结果表明使用进口FSH 2 4mg +PG (Lutalyse○R) 35mg和国产FSH 11mg +PG 0 8mg对水牛超数排卵有效 ,同时提示需要足够数量的受体 ,从中选用发情表现明显、黄体发育良好的进行胚胎移植 ,才能取得良好效果。  相似文献

The cotton whitefly, Bemisia tabaci (Gennadius) B‐biotype, is fed on by a wide variety of generalist predators, but there is little information on these predator–prey interactions, especially under field conditions. In this study, a real‐time polymerase chain reaction (PCR) assay was developed to quantify B. tabaci B‐biotype remains in predator gut. The B. tabaci B‐biotype genomic DNA copy number was referred to the actual amount of BT1 isolate, the B. tabaci B‐biotype specific DNA fragment. The numbers of BT1 isolate in one B. tabaci B‐biotype egg, individual adult and a single red‐eyed nymph were 2.56 × 103, 2.56 × 104, and 1.29 × 104 copies, respectively. When Propylaea japonica adults fed on one, two, four, eight or 16 red‐eyed nymphs, the detected numbers of BT1 isolate ranged from 2.77 × 104 to 4.05 × 105 copies, forming a strong linear relationship (R2 = 0.9899). Following the consumption of two red‐eyed nymphs, prey DNA was detectable in 100% of P. japonica at t = 0, decreasing to 80.0% and 60.0% after 1–4 h and 8 h of digestion, respectively, with 3.36 × 104–1.25 × 103 BT1 isolate copies. The predation by field‐collected predators, 26 larvae of P. japonica, and of Harmonia axyridis each, Chrysopa spp. larvae (Chrysopa pallens and C. formosa, 18 individuals in total), and a single adult of Scymnus hoffmanni, 19 adults of Orius sauteri and nine adult spiders (Erigonnidium graminicolum and Neoscona doenitzi), on B. tabaci B‐biotype were quantified. Of the 99 analysed predator individuals, 3.65 × 102–4.60 × 105 copies of BT1 isolate, equivalent to 0.8–18.8 red‐eyed nymphs were detected. These results suggest that TaqMan real‐time PCR technology may provide a rapid and sensitive method for quantifying B. tabaci B‐biotype remains in predator guts and will be invaluable in assessing the food web relationship between prey and arthropod predators.  相似文献
